Majority Back Call for Cell Phone Ban While Driving

Should Cell Phone Use During Driving Be Completely Banned?With the NTSB calling for a nationwide ban on cell phone use and texting while driving (see last week's iOS News Review), the public has also expressed its opinion on this controversial topic. According to a new poll released by SodaHead.com, claimed to be the Web's largest opinion-based community, 56% of respondents agree with the NTSB and support the ban, while 44% oppose the ban.

When broken down by demographics, 60% of females support the ban and 52% of men are also in favor of the ban. When looking at the results by the age of respondents, the group with the most support for the ban are those over the age of 65 (78%), while the group with the lowest support are those between the ages of 25 and 34 (47%).

Could iPhone 4S Users Get Away with Using Siri Under a No Phones While Driving Ban?

International Business Times' Ranina Sanglap reports that iPhone 4S users who thought they could get away from total bans against using electronic communications devices while driving by using Siri will still be held accountable if federal regulators will have their way.

Sanglap notes that the US National Transportation Board issued a recommendation last week that state governments should ban all driver use of cell phones, handheld or handsfree phones, as well as other portable electronic devices except in cases of emergency, and while the board doesn't have the power to force states to impose the proposed ban, its recommendations do carry significant weight with federal regulators.

The report notes that response to the proposed ban has been largely negative, but in this writer's estimation, the board has made the right call. Research has shown that handheld or handsfree, using a cell phone while driving increases the likelihood of an accident by up to 4x. A study cited by The Coalition for Cell Phone Free Driving says statistical likelihood of being involved in a motor vehicle accident rises by a factor of 4 during cellphone use - greater than with low-level alcohol impairment, and that it slows a driver's reaction time by 18%.

As for the counter-argument that talking on a handsfree phone should be no different than talking to a passenger inside the automobile, that is not the case, a reason being that the person at the other end of a telephone conversation has no sense of what is happening in the car and is unable to react or just shut up immediately should dangerous set of circumstances develop.

An Ontario Medical Association report, based on a meta-analysis of studies from around the world, found talking on a cellphone leads to a substantial reduction in the driver's field of view, changes of driving speed, decreased the distance maintained between vehicles, resulted in more frequent panic braking, less mirror-checking, and slowed response time to traffic light changes.

Sanglap's article cites an analysis by Jim Hedlund, a safety consultant and former National Highway Traffic Safety Administration official, that examined 300 cellphone studies for the Governors' Highway Traffic Safety Administration and concluded that there is little evidence that handsfree mobile phone use is less risky than handheld use, but suggests that the real solution would be more responsible attitudes and self-discipline exercised by drivers.

Apple A5 Chip Production Moves to Texas

The Register's Anna Leach reports that Apple's A5 chips are now being made in Texas, not Asia, as was its predecessor the A4, according to a report from Reuters, citing "people familiar with the operation".

The chips that power the iPad 2 and iPhone 4S are being produced at Samsung's huge new 1.6 million square foot, $3.6bn (closer to $9bn, according to Austin Chamber of Commerce) fab in Austin, although the iOS devices the A5 powers are still assembled by Foxconn in China.

Kindle Fire Missing Manual Editor Going Back to iPad

New York Times blogger David Streitfeld says his NYT article citing high levels of dissatisfaction with Amazon's new tablet generated a torrential response, much of it from people who said they loved their Kindle Fires.

Streitfield notes that some commentators suggested the whole article somehow came from Apple, but none of the conspiracy theorists explained why so many original users of the Fire put mixed-to-negative reviews on Amazon's own site.

Streitfield cites another professional evaluation, from someone who has probably used the Kindle more than anyone who doesn't work for Amazon: Peter Meyers, a digital book consultant and author of Kindle Fire: The Missing Manual, to be published in January as a print volume and an ebook from O'Reilly Media.

He suggests that anyone who would accuse Meyers of bias should consider that if the Fire is a tremendous failure, market for his manual would be negligible.

However, Meyers' verdict, in an email to Streitfield, is that "Apple would have never shipped a device like the Fire. Its got way too many rough edges (sluggish touchscreen, magazine apps that don't really fit the smaller screen, an easy-to-hit power button). And even little things like how the power cord jiggles when plugged in wouldn't have made it past the demo room in Cupertino."

Further, Streitfield says once the manual writing project is finished, Meyers doesn't see much of a future for his own Kindle Fire, which will be going back in the box as soon as he's done, noting that the iPad?2 is "years ahead" of the Fire and lets him "consume and create with no friction."

The Rise of Smartphones, Apps, and the Mobile Web

  • The majority of 25-34 and 18-24 year olds now own smartphones (64% and 53% respectively).
  • The majority of smartphone owners (62%) have downloaded apps on their devices, and games are the top application category used in the past 30 days.
  • The number of smartphone subscribers using the mobile Internet has grown 45% since 2010.
  • 87% of app downloaders (those who have downloaded an app in the past 30 days) have used deal-of-the-day websites like Groupon or Living Social.
  • Younger groups text the most. In Q3, teens 13-17 sent and received the most text messages (an average of 3,417 each month).

This report draws from a broad range of Nielsen data sources, including Nielsen's in-depth monthly surveys of mobile consumers (more than 300,000 consumers surveyed each year); Device metering data from the iOS and Android smartphones of thousands of consumers who have volunteered to be a part of our research panel; detailed, monthly analysis of the cellphone bills for 65,000 lines in the US, again, thanks to volunteer panelists.

iPhone May Preview Personal Computing's Future

Writing for Tech.pinions, Tim Bajarin recalls a 1989 syndicated column he wrote about a modular mobile computing concept, musing about what future portable computing might look like, the the heart of his vision was the idea of having a lot of screens available in his work and home lifestyle, these screens to be displays that his modular computer would connect to in a lot of places and locations, meaning he would always have his own personal computer with him everywhere he went, able to just plug it into an available screen and keyboard.

Bajarin notes that we have modular computing of sorts today in the form of our laptops containing our own OS, customized UI, and all of our personal files that can be plugged into a screen and keyboard as part of our computing model, the way he plugs his MacBook Air to a 27" screen at the office and and uses a wireless mouse and wireless keyboard - the MacBook Air essentially becoming an anchor brick module providing CPU power, OS, UI, and access to files.

"But what if we could have that same kind of modular functionality in a brick that fits in your pocket?" Bajarin asks. "A very small device that houses a powerful CPU, OS/custom UI and data files and can be docked with a multitude of screens that are accessible around the office, school, home, shopping malls, etc. As far out as this seems, I believe that this is exactly the vision Apple has for the future of the iPhone."

Editor's note: It's difficult to dispute that reasoning. I've got the door wedged open a crack what with these days using my three in-service laptops plus an iPad 2 largely as terminals to access and work with a "float" of projects stored on Dropbox. cm

How to Copy Music from Your iPhone, iPad, or iPod touch to Your Computer for Free

Display Shoot-Out: iPad 2 vs. Kindle Fire vs. Nook Tablet

9 to 5 Mac's Christian Zibreg reports on a tablet display shootout among Amazon's Kindle Fire, Apple's iPad 2, and Barnes & Noble's Nook Tablet conducted by Dr. Raymond Soneira, president of DisplayMate Technologies Corporation of Amherst, N.H.

Dr. Soneira set out to find out which device packed the best IPS (In-Plane Switching) display. Zibreg points out that LG Displays, the world's leading manufacturer of IPS LCDs, is the supplier of IPS panels for the three devices used in the shoot-out, but the iPad?2, the test note said, remains the gold standard in tablet displays, while Motorola's Xoom and Acer's Iconia Tab serve as prime examples of low-quality LCD displays.

The report notes that the Fire's display is on par with iPad 2's and Nook Tablet's in most respects, except for two important aspects - limited grayscale rendering and an antireflective treatment with about 70% higher reflectance levels compared to the iPad?2 and more than double the reflectance of the Nook Tablet - bad for reading outdoors or in brightly lit indoor areas. Meanwhile, Barnes & Nobles Nook Tablet has the lowest reflectance of any tablet DisplayMate ever tested, showing 28% lower reflectance than even the iPad 2, and grayscale rendering more accurate than most living room HDTVs.

Consequently, Nook wins the shootout.

iPad Keyboard Showdown

Laptop Mag's Michael A. Prospero observes that the iPad's virtual keyboard is definitely good, but nothing feels quite as nice as clickety-clacking away on a physical keyboard. According to Lenovo and Qualcomm, 71% of small businesses surveyed said they are using tablets at work as an additional device, not as a replacement for their PCs. But with the right case, complete with a physical QWERTY Bluetooth keyboard, that might be about to change.

Many accessory manufacturers are ready to step in with external keyboards, but which one offers the best combination of price, portability, protection, and tactile feedback? Make the Most of iOS 5's Hidden Features on the iPad

APCMag's Dan Warne notes that while iOS 5 has a bundle of great headline features, it also packs in a few extremely useful ones that you might not even notice.

There is a thumb-friendly split keyboard for your iPad, plus custom autotext shortcuts, which can be extremely useful for saving repetitive typing of things you often add in to emails, instant message chats or documents, but unfortunately hidden away in a submenu that very few people would ever look at (go to Settings > General > Keyboard > Add new shortcut).

Then there's multitouch app switching. Warne observes that one of the biggest time-wasters when working with a tablet is switching between apps, but iOS 5 introduces a multitouch method of app-switching: the four finger swipe, plus a five finger pinch to close the app and return to the home screen.

Rumor: Apple to Launch 7.85" iPad in 2012

DigiTimes' Max Wang and Steve Shen report that Apple is likely to launch a 7.85" iPad prior to the fourth quarter of 2012 in addition to a new iPad scheduled to be released at the end of the first quarter, according to sources in the supply chain.

Wang and Shen note that global shipments of tablet PCs are expected to reach 60 million units in 2011, of which 70% will be Apple's iPads. However, their insider sources tell them that in order to stay in front of increasing market competition, including the 7" Kindle Fire from Amazon and the launch of large-size smartphones from handset vendors, Apple has been persuaded to develop 7.85" iPads, with OEMs in the supply chain, including panel makers LG Display and AU Optronics (AUO), likely to begin production of the 7.85" models at the end of Q2 2012.

Fitch Rates Florida's $239MM GO Full Faith & Credit Right-of-Way Bonds 'AAA'; Outlook Negative

NEW YORK--(BUSINESS WIRE)--Fitch Ratings assigns an 'AAA' rating to the following state of Florida full faith and credit bonds:

--$238.77 million Department of Transportation right-of-way acquisition and bridge construction bonds, series 2011B.

The bonds are expected to sell competitively as soon as the week of Jan. 3 for bids on 18 hours' notice.

In addition, Fitch affirms the following ratings:

--Approximately $14 billion in outstanding Florida full faith and credit bonds at 'AAA'.

The Rating Outlook is Negative.

KEY RATING DRIVERS

SOLID LONG-TERM ECONOMIC PROSPECTS: Long-term economic prospects are solid, although current economic performance remains weak. Economic fundamentals are strong with future growth expected; however, the housing market remains weak and the unemployment rate above average.

STRONG FINANCIAL MANAGEMENT PRACTICES: The state employs sound financial management practices, including the use of consensus revenue estimating and a history of prompt action to maintain fiscal balance.

SIGNIFICANT RESERVES: Florida has maintained large reserves, totaling more than $3.46 billion as of fiscal 2011 year-end. These reserves offset risks associated with an economically sensitive revenue system that is vulnerable to declines in the rates of population growth, consumption, and activity in the housing market.

MODERATE LIABILITIES: The state's debt burden is moderate and pensions are well funded.

WHAT COULD TRIGGER A RATING ACTION

--Inability to maintain budget balance while preserving an adequate reserve position;

--Economic performance weaker than what is assumed in state forecasts.

SECURITY

Florida's full faith and credit bonds are secured first by specific revenues, in this case, a first lien on motor and diesel fuel taxes. Florida's full faith and credit are also pledged and provide the basis for the rating.

CREDIT PROFILE

Florida's 'AAA' general obligation (GO) rating recognizes the state's strong financial management practices, moderate debt burden, well-funded pension system, solid long-term economic prospects, and still significant reserves, including various trust funds. The Negative Outlook reflects the severity of the state's economic and revenue decline as well as continuing uncertainty associated with the economic and revenue outlook.

Until the recent recession, the Florida economy was characterized by rapid growth, economic broadening and diversification as it was transformed from a narrow base of agriculture and seasonal tourism into a service and trade economy, with a substantial insurance, banking and export components. Strong underlying fundamentals remain, including a relatively low cost of living, attractive tourist and retirement destinations, and favorable geographic location; however, there is significant uncertainty regarding the near term economic outlook and economic performance during the recession was among the weakest of the states. The state's natural amenities include 2,200 miles of tidal shoreline, proximity to Latin American and Caribbean markets, the presence of some of the world's most popular tourist destinations, large convention venues, and major cruise ship ports.

Florida's poor economic performance in the downturn, one of the most negative among the states, reflects the state's severe housing market correction following the historic run-up. State employment was down 3.5% in 2008, compared to a 0.6% loss for the nation, another 6.2% in 2009, much worse than the 4.4% loss for the U.S., and a further 1.1% in 2010, when the U.S. declined 0.8%. Year-over-year performance has been improving and state employment has grown each month since the start of calendar 2011, the first year-over-year growth since June 2007. Employment growth had generally been lagging the national rate although November non-farm employment increased 1.4% year-over-year, surpassing the U.S. for the third consecutive month. Construction employment continues to drop, albeit at a slower pace, and is less than half what it was in 2006. The state's unemployment rate, although down from a historical high of 12% in December 2010, was 116% of the U.S. rate at 10% in November 2011.

Florida's revenue sources (primarily a sales tax but also a documentary stamp tax in large part based on real estate transactions) have been especially susceptible to the state's steep housing market correction; the state has no personal income tax. The Florida legislature consistently and promptly addressed numerous large negative revenue estimate revisions during the downturn, maintaining budget balance and an adequate reserve position. A large budget gap for fiscal 2010 was closed primarily through the use of federal stimulus monies and some trust fund balances, spending reductions, and significant fee and tax increases ($1.9 billion, including about $850 million from a $1/pack cigarette tax increase). Except for those increases, the state has not relied on revenue measures to close budget gaps. The budget for fiscal 2011 addressed a gap of about $2.8 billion with federal stimulus funds, reserve draws, spending control, and $433 million from a gaming compact with the Seminole Tribe.

The enacted fiscal 2012 budget, which totals $69.2 billion, a reduction of $1.5 billion (2.1%) from the fiscal 2011 budget, closed a projected $3.6 billion gap with spending reductions and a requirement that employees begin making contributions to retirement plans. The budget, as enacted, is structurally balanced and does not use non-recurring revenues to support on-going spending. It is projected to generate an operating surplus that would be used to add to general fund and budget stabilization reserves.

Reserves at fiscal 2011 year-end remained significant with $1.03 billion (unaudited) in general fund reserves, including the budget stabilization fund of $281.3 million, and trust fund balances of $2.4 billion, including $769 million in the Lawton Chiles Endowment Fund. Although aggregate reserves are sharply reduced from the peak of $9.9 billion in fiscal 2006 and down from original expectations for the year, the state's maintenance of a solid reserve position in such a strained financial environment has been notable and a key credit strength. The fiscal 2012 budget as enacted would add to reserves and begin the rebuilding of the budget stabilization fund as required by law after a drawdown.

The state's slow economic recovery, however, continues to have an effect on revenues as is indicated by a $600 million (2.5%) downward revision in expected revenues for fiscal 2012 as of the October revenue estimating conference. This revenue shortfall is expected to be accommodated through the use of general reserves, which were $289 million higher than anticipated during budget preparation. General Fund revenue is still expected to grow 2.9% on a year-over-year basis. Revenues through October were 0.2% above estimate and 2.2% higher year-over-year. The governor's proposed budget for fiscal 2013 would further reduce appropriations to $66.4 billion and increase allocations to education while reducing spending on Medicaid and corrections. Per pupil education funding would rise 2.3% to $6,372 from the current $6,230, still well below the fiscal 2008 peak of $7,126.

The state's debt position and structure are conservative. Debt represents a moderate burden on Florida's resources with net tax-supported debt of about $23.6 billion equal to 3.2% of personal income. Florida's debt portfolio does not include derivatives and variable-rate debt is negligible at less than 0.5% of net tax-supported debt. Pensions had been overfunded since fiscal 1998, but due to market losses and assumption changes to reflect the results of a 2009 experience study the funded ratio dropped to a still solid 87% as of July 1, 2010.

Florida's full faith and credit bonds are secured first by specific revenues. The right-of-way acquisition and bridge construction bonds are payable primarily from pledged motor and diesel fuel taxes that are transferred from the state transportation trust fund (STTF) to the right-of-way acquisition and bridge construction fund, and ultimately by Florida's full faith and credit pledge. State law limits the amount to be transferred from the STTF to the lesser of 7% of STTF revenues or $275 million. Estimated fiscal 2012 pledged revenues provide 7.5x coverage of pro forma debt service and 5.8x coverage of maximum allowable debt service. Minimum maximum annual debt service (MADS) coverage of 1.11x by pledged gas taxes is required for parity bond issuance. Consumption of and tax receipts resulting from motor and diesel fuels have grown consistently and tax rates are increased annually with the consumer price index.

Back in campaign mode, presidential hopefuls focus (AP)

DES MOINES, Iowa ? Republicans in search of their party's presidential nomination are returning to campaign mode after a brief Christmas respite, with Rick Santorum planning a hunting trip with conservatives in Iowa and Mitt Romney phoning supporters.

With just a week until Iowa holds its leadoff caucuses and many caucus-goers undecided, the final push ahead of the Jan. 3 contests was heading into a critical time. Campaigns planned new television ads and phone calls to persuade holdouts still weighing their options.

Romney, who kept this state at arm's length for most of the year, seemed to increase his efforts in Iowa as polls found him in a stronger position. He planned to talk with supporters in a series of telephone calls here and to New Hampshire and Florida on Monday between working on a speech that aides described as his final pitch to Iowans. Romney planned to deliver that speech Tuesday evening and then set out on a bus tour of Iowa.

However, he was to share the highways with Rep. Michele Bachmann, Texas Gov. Rick Perry and former House Speaker Newt Gingrich. All scheduled bus tours to start then, too.

Each is running out of time and looking to derail Rep. Ron Paul, the Texas congressman who seems to have the most sophisticated network of volunteers ready to organize for the caucuses. Paul was to return to Iowa this week to meet with supporters he has kept in touch with since his unsuccessful run in 2008.

Others, too, were ready to turn on their own political machines and put fresh ads on the air.

Gingrich, who last week criticized the negative tone of the campaign, was preparing to directly challenge Romney on the economy, an issue Romney has made central to his campaign. Gingrich's standing in public and private polls has slipped as he faced unrelenting criticism from the candidates and their allies.

Gingrich was expected to use clips from Romney's previous campaigns distancing himself from President Ronald Reagan and pitch his own economic plan as "Reaganomics 2.0." Gingrich also was expected to compare Romney's tax plan with his own.

Romney released a new TV ad in Iowa on Monday that touts him as a conservative businessman and in which he says "it is a moral imperative for America to stop spending more money than we take in."

"It's killing jobs and it's keeping our kids from having the bright prospects they deserve," he says in the spot. "The experience of balancing budgets is desperately needed in Washington and I will take it there."

Santorum, meanwhile, planned to announce support from another wave of Iowa conservatives. He scheduled a pheasant hunting trip in Adel for Monday afternoon. While he trails in polls and has not spent significant money on ads, Santorum is hoping his nonstop courtship of Iowans yields a late surge. He visited all 99 of Iowa's counties during the summer ? an accomplishment Bachmann has feverishly tried to replicate.

Bachmann, a congresswoman from Minnesota, last week darted through small towns, reminding voters that Santorum lost his 2006 re-election bid in a blowout and that Paul's foreign policy views were outside the party's orthodoxy.

Looking to recapture voters' interest, her plan was to return to hand-to-hand campaigning Tuesday and paint herself as the only acceptable conservative in the race.

"You can always count on me to stay true to my word and put America and Americans first," she said in a fundraising email sent Monday. "I am the consistent conservative who will put our nation back on a path towards prosperity and restore our values to government."

Perry, too, was looking to keep up the message that his rivals are insiders unable to change Washington. He planned to resume his tour bus on Tuesday.

Former Utah Gov. Jon Huntsman kept his focus on New Hampshire. Huntsman said early in the campaign that he would not compete in Iowa and instead make his start in New Hampshire, which comes second on the nominating calendar with a Jan. 10 primary.

Obama marks Christmas with gifts, carols, church (AP)

HONOLULU ? President Barack Obama exchanged gifts with his family, sang carols and attended church services as he celebrated Christmas in Hawaii.

The president and his family woke up early Sunday to exchange gifts, the White House said, then had breakfast and sang Christmas carols at the multimillion-dollar house they rent in Kailua Beach, near Honolulu.

Later in the morning, the Obamas made the short trip to the chapel at Marine Corps Base Hawaii for Christmas church services. The president dressed casually in dark khaki pants and a short-sleeve blue shirt, and his wife and daughters donned sundresses for Christmas services on a bright and breezy day on the island of Oahu.

The White House said the president and Michelle Obama would return to the base later in the day to visit with service members and their families, as they have done in past years. Many of the Marines stationed at the base have deployed to Afghanistan, as well as Iraq, where the last American troops were withdrawn earlier this month.

The president also called 10 service members stationed around the world ? two from each branch of the military ? on Christmas Eve. The White House said he thanked them for their service and the sacrifice of being away from their families at the holidays.

The Obamas planned to wrap up their Christmas festivities with dinner at the rental home with friends and family. Among those joining the first family in Hawaii are the president's sister, Maya Soetoro-Ng, who lives on Oahu, and several friends Obama has known since high school.

The president has kept a low profile since arriving in Hawaii on Friday evening to start a vacation delayed by the stalemate in Washington over extending payroll tax cuts. He has no public events planned, and his only outings are expected to be to the golf course or to take his daughters for shave ice, a Hawaiian snow cone.

The Obamas are expected to return to Washington shortly after New Year's Day.

East Chicago Mayor Copeland Threatened

East Chicago, Ind. - East Chicago, Ind. Mayor Anthony Copeland was threatened by a gunman on his Thursday night walk.

The incident occurred at 10 p.m. next to Johnny?s Meat Market, 3940 Main St., near Copeland?s home.

?I usually go out for a nightly two-mile walk,? Copeland said. ?I used to do it during the day. I was coming back home, when about a block and half from my house, a guy came out of the alley and tried to rob me.?

Copeland said he didn?t break stride as he took cover behind a truck parked on the street. The gunman never broke the cover of the alley, Copeland said.

?I yelled at the top my lungs, ?Are you going to shoot me or what are you going to do?,? ? Copeland said.

The man never responded and instead fled back down the alley.

Copeland called police, and they searched for the man to no avail. The suspect was described as a black male in his 30s, wearing black jeans, a black jacket, a black hooded sweatshirt and a black knit hat. Copeland hurt his shoulder as he took cover behind the truck.

A background in self-defense may have helped Copeland avoid serious injury. Copeland taught self-defense classes for 15 years, and one of the techniques has stayed with him in the years since.

?When I walk and come to alley or any kind of entryway, I always instinctively veer away from it by about five or six feet, in case a car or dog comes out,? Copeland said. ?If I had stayed alongside the building, he would have pointed it at my head or body.?

Copeland said he doesn?t think the man knew who he was. ?It was just a thing of chance and opportunity,? Copeland said.

The situation did leave Copeland a bit shaken, but he?s determined not to change his routine.

?What am I going to be ? a hermit inside my home?? Copeland said. ?It alters you. Will I think twice about taking the route again? Yes. Will I be paralyzed in fear? No.

?Only a foolish person would think it couldn?t happen to them. Any person, any time, any place ? that person could take your life from you. I know that I?m overwhelmed with blessings.?

Computer assisted design (CAD) for RNA

ScienceDaily (Dec. 22, 2011) ? The computer assisted design (CAD) tools that made it possible to fabricate integrated circuits with millions of transistors may soon be coming to the biological sciences. Researchers at the U.S. Department of Energy (DOE)'s Joint BioEnergy Institute (JBEI) have developed CAD-type models and simulations for RNA molecules that make it possible to engineer biological components or "RNA devices" for controlling genetic expression in microbes. This holds enormous potential for microbial-based sustainable production of advanced biofuels, biodegradable plastics, therapeutic drugs and a host of other goods now derived from petrochemicals.

"Because biological systems exhibit functional complexity at multiple scales, a big question has been whether effective design tools can be created to increase the sizes and complexities of the microbial systems we engineer to meet specific needs," says Jay Keasling, director of JBEI and a world authority on synthetic biology and metabolic engineering. "Our work establishes a foundation for developing CAD platforms to engineer complex RNA-based control systems that can process cellular information and program the expression of very large numbers of genes. Perhaps even more importantly, we have provided a framework for studying RNA functions and demonstrated the potential of using biochemical and biophysical modeling to develop rigorous design-driven engineering strategies for biology."

Keasling, who also holds appointments with the Lawrence Berkeley National Laboratory (Berkeley Lab) and the University of California (UC) Berkley, is the corresponding author of a paper in the journal Science that describes this work. The paper is titled "Model-driven engineering of RNA devices to quantitatively-program gene expression." Other co-authors are James Carothers, Jonathan Goler and Darmawi Juminaga.

Synthetic biology is an emerging scientific field in which novel biological devices, such as molecules, genetic circuits or cells, are designed and constructed, or existing biological systems, such as microbes, are re-designed and engineered. A major goal is to produce valuable chemical products from simple, inexpensive and renewable starting materials in a sustainable manner. As with other engineering disciplines, CAD tools for simulating and designing global functions based upon local component behaviors are essential for constructing complex biological devices and systems. However, until this work, CAD-type models and simulation tools for biology have been very limited.

Identifying the relevant design parameters and defining the domains over which expected component behaviors are exerted have been key steps in the development of CAD tools for other engineering disciplines," says Carothers, a bioengineer and lead author of the Science paper who is a member of Keasling's research groups with both JBEI and the California Institute for Quantitative Biosciences. "We've applied generalizable engineering strategies for managing functional complexity to develop CAD-type simulation and modeling tools for designing RNA-based genetic control systems. Ultimately we'd like to develop CAD platforms for synthetic biology that rival the tools found in more established engineering disciplines, and we see this work as an important technical and conceptual step in that direction."

Keasling, Carothers and their co-authors focused their design-driven approach on RNA sequences that can fold into complicated three dimensional shapes, called ribozymes and aptazymes. Like proteins, ribozymes and aptazymes can bind metabolites, catalyze reactions and act to control gene expression in bacteria, yeast and mammalian cells. Using mechanistic models of biochemical function and kinetic biophysical simulations of RNA folding, ribozyme and aptazyme devices with quantitatively predictable functions were assembled from components that were characterized in vitro, in vivo and in silico. The models and design strategy were then verified by constructing 28 genetic expression devices for the Escherichia coli bacterium. When tested, these devices showed excellent agreement -- 94-percent correlation -- between predicted and measured gene expression levels.

"We needed to formulate models that would be sophisticated enough to capture the details required for simulating system functions, but simple enough to be framed in terms of measurable and tunable component characteristics or design variables," Carothers says. "We think of design variables as the parts of the system that can be predictably modified, in the same way that a chemical engineer might tune the operation of a chemical plant by turning knobs that control fluid flow through valves. In our case, knob-turns are represented by specific kinetic terms for RNA folding and ribozyme catalysis, and our models are needed to tell us how a combination of these knob-turns will affect overall system function."

JBEI researchers are now using their RNA CAD-type models and simulations as well as the ribozyme and aptazyme devices they constructed to help them engineer metabolic pathways that will increase microbial fuel production. JBEI is one of three DOE Bioenergy Research Centers established by DOE's Office of Science to advance the technology for the commercial production of clean, green and renewable biofuels. A key to JBEI's success will be the engineering of microbes that can digest lignocellulosic biomass and synthesize from the sugars transportation fuels that can replace gasoline, diesel and jet fuels in today's engines.

"In addition to advanced biofuels, we're also looking into engineering microbes to produce chemicals from renewable feedstocks that are difficult to produce cheaply and in high yield using traditional organic chemistry technology," Carothers says.

While the RNA models and simulations developed at JBEI to date fall short of being a full-fledged RNA CAD platform, Keasling, Carothers and their coauthors are moving towards that goal.

"We are also actively trying to make our models and simulations more accessible to researchers who may not want to become RNA control system experts but would nonetheless like to use our approach and RNA devices in their own work," Carothers says.

While the work at JBEI focused on E. coli and the microbial production of advanced biofuels, the authors of the Science paper believe that their concepts could also be used for programming function into mammalian systems and cells.

"We recently initiated a research project to investigate how we can use our approach to engineer RNA-based genetic control systems that will increase the safety and efficacy of regenerative medicine therapies that use cultured stem cells to treat diseases such as diabetes and Parkinson's," Carothers says.

UPDATE 1-French police to ease Christmas airport strike

France has ordered police to deploy at two airports from Thursday to make sure striking private security staff do not paralyse flights in the pre-Christmas travel rush, a police union official said on Wednesday.

President Nicolas Sarkozy's government had said it was ready to order police to take over security at Lyon airport and part of Charles de Gaulle, the busy international hub on the northern outskirts of Paris.

"If the strike continues, the government will see to it that normal transport service is guaranteed," Transport Minister Thierry Mariani said. "We cannot yet again have a situation where the French people are prevented from going home to their families."

About 400 police were put on standby on Wednesday after the pay talks broke up without agreement. The talks were due to resume on Thursday but a police union official said the police had been instructed to intervene from Thursday.

The Interior Ministry would only say that "the operation is in place and ready to be activated at any moment".
